VECLADX (venetoclax) is used for the treatment of adult patients with:
- Chronic lymphocytic leukemia (CLL) — as fixed-duration therapy in combination regimens (e.g., with obinutuzumab or rituximab) or as monotherapy in relapsed/refractory disease, including 17p deletion / TP53-mutated CLL.

Venetoclax selectively binds the anti-apoptotic BCL-2 protein that CLL cells and AML blasts rely on for survival. Dosage & Administration

For CLL, venetoclax is initiated with a 5-week once-daily ramp-up (20 mg → 50 mg → 100 mg → 200 mg → 400 mg) with mandatory tumor lysis syndrome (TLS) prophylaxis, hydration and laboratory monitoring, then continues at 400 mg once daily. For AML in combination with azacitidine, ramp up 100 mg → 200 mg → 400 mg over cycle 1 days 1–3, then 400 mg daily. Tablets are taken orally with food and water, swallowed whole, at the same time each day. Treatment must be initiated and supervised by an experienced hematologist/oncologist.
Side Effects
The most clinically important risk is tumor lysis syndrome (TLS) during initiation in high-tumour-burden patients. Common adverse reactions include neutropenia (with infection risk), diarrhoea, nausea, anaemia, thrombocytopenia, fatigue and cough. Strong CYP3A4 inhibitors/inducers require dose adjustment or avoidance. Venetoclax can cause fetal harm; effective contraception is required during and after treatment.
