This broad action blocks both the growth signals inside cancer cells and the signalling that drives new blood-vessel formation, so tumours are deprived of both the instructions to divide and the blood supply they need to survive and spread.

The three approved indications are advanced colorectal cancer after standard chemotherapy, imatinib- and sunitinib-refractory gastrointestinal stromal tumour, and sorafenib-refractory hepatocellular carcinoma. Dosage
The recommended dose is 160 mg (four 40 mg tablets) once daily with a low-fat meal, given in 28-day cycles of 21 days on and 7 days off. A 28-tablet pack covers seven days of the active-treatment phase. The dose may be reduced to 120 mg or 80 mg daily to manage side effects such as hand-foot skin reaction or elevated liver enzymes.
Side Effects
The most frequent side effects are hand-foot skin reaction, diarrhoea, fatigue, high blood pressure, reduced appetite and mouth sores. Serious but uncommon effects include liver injury, bleeding, gastrointestinal perforation and cardiac problems. Liver function, blood pressure and blood counts should be monitored regularly, and patients should report skin changes, persistent diarrhoea, dark stools or chest pain promptly.
