In BRAF-mutant tumors the MAPK pathway is permanently active, driving cancer cells to grow and survive. Trametinib binds MEK1 and MEK2 allosterically and silences them, interrupting that growth signal downstream of the mutated BRAF. Adding dabrafenib blocks the pathway at two separate points, which deepens and prolongs the response and helps delay the emergence of resistance.

Dosage & Administration

The standard dose is 2 mg once daily, taken at the same time each day with or without food. When combined with dabrafenib, Mekinist is taken once daily while dabrafenib is taken twice daily. Tablets are swallowed whole. The oncologist may reduce the dose or hold treatment if side effects arise.

Side Effects

Rash, diarrhea, fatigue, nausea, swelling of the limbs and fever are the most common effects. More serious risks include reduced heart function (cardiomyopathy), blood clots, severe skin reactions, eye problems such as retinal vein occlusion, lung inflammation, and bleeding. Heart function and skin and eye symptoms are monitored during therapy.
