Generic Information: Lumakras (Sotorasib) — Amgen US

Lumakras is the brand name for sotorasib, the first medicine ever approved to target the KRAS G12C mutation. For more than three decades KRAS was written off as "undruggable" because its surface offered no obvious pocket for a drug to grip. Amgen cracked that problem by designing a molecule that latches onto a cysteine amino acid unique to the G12C mutant, and Lumakras became the first such inhibitor cleared by the U.S. FDA. The U.S.-market Lumakras is supplied in 120 mg tablets, with larger 240 mg and 320 mg presentations available to suit different dosing and supply needs.

What Is Lumakras (Sotorasib) Used For?

Lumakras treats cancers driven by the KRAS G12C mutation. Its licensed indications are:

  • KRAS G12C-mutated non-small cell lung cancer (NSCLC) that is locally advanced or metastatic, in adults who have had at least one prior systemic therapy.
  • KRAS G12C-mutated metastatic colorectal cancer (mCRC).

Because it is a targeted drug, it acts on tumour cells carrying the specific mutation and leaves most healthy tissue alone.

How Does Lumakras (Sotorasib) Work?

KRAS is a signalling protein that flips between an active "on" and an idle "off" state. In healthy cells this switch is tightly controlled, but the G12C mutation jams it in the "on" position so the cell keeps dividing without a brake. Sotorasib binds permanently to the mutant's cysteine residue, trapping KRAS in the off state and cutting the growth signal at its source. Starved of this signal, KRAS G12C-driven tumours shrink or stop growing.

Indications

  • KRAS G12C-mutated locally advanced or metastatic NSCLC after prior systemic therapy
  • KRAS G12C-mutated metastatic colorectal cancer

Dosage & Administration

The standard Lumakras dose is 960 mg once daily — that is eight 120 mg tablets swallowed whole at the same time each day, with or without food. Treatment carries on until the cancer progresses or side effects become unacceptable. Your oncologist may pause or lower the dose to manage adverse reactions, so follow their instructions rather than adjusting on your own.

Side Effects

The reactions reported most often are diarrhoea, nausea, vomiting, tiredness, muscle or joint pain, reduced appetite, cough and a rise in liver enzymes. The serious risks that require vigilance are liver injury, interstitial lung disease or pneumonitis, and QT prolongation. Liver blood tests and any new breathlessness or cough should be checked regularly throughout treatment.

Storage & Handling

Keep Lumakras tablets in the original container at room temperature (20–25 °C), shielded from moisture and light. Store out of children's reach and do not use past the expiry date.
