What Is Revolade?

Revolade is an oral platelet-raising therapy. It can also support interferon-based hepatitis C treatment by correcting thrombocytopenia.

How Does Revolade Work?

Eltrombopag binds the transmembrane region of the thrombopoietin receptor c-Mpl on bone-marrow megakaryocytes, triggering JAK2/STAT signalling that stimulates megakaryocyte growth and platelet production. Because it occupies a site distinct from the endogenous thrombopoietin binding site, its effect adds to — rather than competes with — the body's natural TPO.

  • Chronic ITP (adults and children ≥6 years) refractory to corticosteroids or IVIG
  • Severe aplastic anemia refractory to immunosuppressive therapy
  • Thrombocytopenia in chronic hepatitis C to facilitate interferon therapy

Dosage

Adults with ITP typically begin at 50 mg once daily; East Asian patients and those with liver impairment start lower. The dose is adjusted to keep platelets at or above 50 × 10⁹/L, not exceeding 75 mg daily in ITP. Take on an empty stomach, one hour before or two hours after food, and two hours apart from calcium- or mineral-rich meals and supplements.

Side Effects

Common effects are headache, nausea, diarrhoea and fatigue. Serious risks include hepatotoxicity (warranting regular liver function monitoring), thrombosis, and cataract. Platelet counts and liver enzymes should be monitored throughout treatment.
